临床前成像(PCI)对于理解身体在健康和疾病状态下如何工作以及描述对生理或环境变化的响应具有重要作用,它能够在器官、组织、细胞和分子水平提供关于疾病原理的重要见解。这些知识指导着新型治疗策略的开发,而后者能够改善患者预后、挽救生命。
2018 年 12 月 16 日,2018 复旦科技创新论坛在上海成功举办,布鲁克 PCI 部门全球总裁 Wulf-Ingo Jung 博士受邀参加了本次大会,并在 2018 功能与高分子影像分论坛上发表了题为《超高场下小动物磁共振成像的研究进展:技术与应用》的精彩演讲。丁香园有幸邀请到 Jung 博士,就 PCI 在生物医疗领域的价值进行阐述。

PCI 在全球的发展和应用现状
Jung 博士指出,PCI 所涉及的范围非常宽广,从基础科学到应用科学,从生物学、化学、物理学、 医学到他们的交叉学科等等;如今在全球范围内,PCI 的应用价值日益增高。以神经领域为例,该领域的医学技术进展高度依赖于 PCI 的科研成果,尤其是对于许多在老龄化社会中占比很高的神经功能退行性疾病,利用专门的尖端 PCI 仪器能够更快地将科研成果转化为临床诊断应用中。
正如 Jung 博士在大会演讲中所介绍的,超高场 MRI 是布鲁克所能够提供的尖端 PCI 仪器之一。而所谓超高场 MRI,是以更高的场强进行扫描,高磁场强度具有提高信噪比和分辨率以及减少测量时间等独特优势。
Jung 博士表示,据预测,达到 20 特斯拉的磁共振成像将在临床领域打开人类疾病的全新视角,虽然有人认为距离临床实现 20 特斯拉 MRI 的应用可能尚需要耗费 10~20 年之久,但目前布鲁克应用于 PCI 的 MRI 已经实现了这一场强,也让该领域各项前沿的临床前研究有了得以开展的可能。
除了前述超高场 MRI 等更尖端的仪器的研发,目前 PCI 发展的另一个主要趋势是多模式成像,即将不同的成像技术组合成一种仪器。Jung 博士指出,这是因为多模式成像能够将不同成像模式之间的优点整合,在科研和临床方面提供更准确、全面的图像信息,助力研究人员在生物医学领域的项目获得成功。
应用领域广、技术领先的布鲁克 PCI 解决方案
Jung 博士指出,布鲁克能够为广泛的应用领域提供先进的 PCI 解决方案,例如,癌症研究、功能与解剖神经成像、整形外科、心脏成像和卒中模型。
比如,在高达 18 特斯拉的布鲁克超高场 MRI 帮助下,研究人员能够深入开展高难的神经科学研究,甚至实现许多在以往看来不可能实现的任务,如「活体 MRI 组织解剖」。此外,布鲁克的显微 CT 产品非常适合用来帮助研究人员深入了解肺部和骨骼疾病,并且已延伸到了牙科研究和其他领域。PET 示踪剂的开发也是布鲁克的一个非常重要的研究课题,其核分子成像(NMI)产品具有极高灵敏度和全视野高精度的优势,为药物研发提供有力的工具,通过将 PET 提供的高敏感代谢信息与 MRI 提供的形态学信息结合起来,除了提供解剖学背景外,MRI 还可以进一步研究肿瘤的微观结构,因此 PET/MR 可以深入了解肿瘤的进展和治疗。

首先,布鲁克是全球知名的可以提供 11.7 特斯拉和高达 18 特斯拉超高场临床前 MRI 系统的供应商,极大地推动了目前临床前研究的可能。除此之外,所提供的 MRI 冷冻探头,与传统的室温探头相比,该冷冻探头可大大提高 2~5 倍灵敏度,另外如果没有低温探头,很多高端应用是不可能实现的。
此外,借助 Paravision360 成像操作软件,布鲁克引入了一个无缝集成多模式工作流的新软件平台(Paravision360 提供了方便的优化程序,从动物定位扫描程序,到数据处理和管理),此工作流概念是保证数据准确性和再现性的基础,可实现量化的一致性,以促进全局发现和验证过程。而借助 Labscape 售后平台,布鲁克还能够为客户提供全球服务网络支持下的完整的服务解决方案。
布鲁克致力于成为中国 PCI 市场领军企业
Jung 博士认为,中国科研成像用户对 7 和 9.4 特斯拉高场 MRI 很感兴趣,在许多领域(CEST、高分辨率功能磁共振成像、SWI、杂核成像、磁共振光谱等)都显示出了强大的需求,非人灵长类的脑研究也极大促进了大口径动物 MRI 的研发,这为生物医学研究领域新的突破性发现开辟了独特的可能性。此外,中国对 NMI 和 UCT 产品日益增长的需求也证实了多模成像的附加价值。
创立 55 年来,布鲁克一直坚定地致力于满足客户的深层次需求,其不仅是细胞生物学、临床前成像、临床表型组学与蛋白质组学研究、临床微生物学及分子病理学研究领域的高性能系统供应商,同时也提供从应用技术到维护服务的网络支持,持续不断地为当今的分析难题开发最先进的技术和创新解决方案。
Jung 博士表示,若灵长类的治疗策略在未来得到验证,中国将成为一个医疗科研和新药研发的最核心的地方。因此,布鲁克正投入开发一种新的磁共振仪器,应用于非人类灵长类动物的研究。除此之外,布鲁克也持续增加在华投资,不断培养和塑造专业人才,为中国营销市场和科研界提供各方面支持。
访谈最后,Jung 博士强调,中国市场是布鲁克公司战略发展的关键市场,而布鲁克将把握这一发展机遇,进一步加大在华投资力度,努力将自身打造成为中国 PCI 市场的领军企业,希望成为中国临床前成像学界的首选合作伙伴。

Dr Wulf-Ingo Jung, the global president of Bruker PreClinical Imaging (PCI) division, was invited to deliver the Ultra High Field MRI keynote speech in the Fudan science and innovation Forum on 16th Dec 2017 in Shanghai. It is a great honor to invite Dr Jung to give us an interview in Dingxiangyuan about High field MRI impact on BioMedical research field.
Q1 Reporter: Dr Jung, Welcome to Shanghai. At Fudan university, I am impressive to see the highest field MRI, Bruker 11.7T, in China. As a senior expert in MRI, could you please comment on the current status of the development and application of preclinical imaging (PCI) in the world?
Dr Jung: Thanks. Preclinical imaging plays an important role in fundamental, bio-medical and pharmacological research worldwide. Today, dedicated cutting edge preclinical instruments enable faster translation into the clinics.
E.g. brain function is one important field of preclinical research. Progress in this field is highly dependent on preclinical imaging with many applications reaching into neurogenerative diseases from which our aging population is suffering more and more.
For Ultra-High Field MRI preclinical imaging is even spear heading the field with even higher field strengths with the unique advantages like increased SNR and resolution and reduced measurement times. It is speculated that ca. 20T would open completely new insights in human disease – in the preclinical field this field strength is available today! We need to leverage this as waiting for human 20T magnets may need one or two decades!
Currently one major trend we see in preclinical imaging is multimodal imaging: The combination of different imaging technologies into one instrument. Thus, the advantages, benefits and applications of the different modalities complement each other in a single instrument to provide the full picture.
Q2 Reporter: Imaging is the very important modality in different research areas, from basic science to applied science, from biology, chemistry, physics, medicine to their interdisciplinary and so on. So what are the main application areas of Bruker PCI products at present?
Dr Jung: Since Bruker offers a wide multimodal preclinical portfolio our customers apply our systems successfully in all the mentioned areas and more.
To be more specific one main research area of our customers is neurology, for example when using our dedicated MRI systems. With the possibility of ultra high fields of up to 18 Tesla researchers are enabled to perform applications which were not feasible before such as “in vivo MRI microscopy”.
On the other hand, our micro-CT products help researchers to understand lung and bone diseases since this technology is ideally suited for these applications. This extends into dental research, and other fields, too.
Another essential and very important research topic is PET tracer development for which our Nuclear Molecular Imaging (NMI) products are ideally suited with their superior sensitivity and their full FOV accuracy.
One common main application is oncology. Specifically, the combination of the highly sensitive metabolic information provided by PET together with morphological information from MRI is very beneficial. Besides providing the anatomical context, MRI allows also to further investigate the tumor micro-structure and thus Bruker PET/MR allows for deep understanding of the tumor progression and treatment.
Q3 Reporter: Compared with other companies, what are the main competitive advantages of Bruker PCI in the market?
Dr Jung: Bruker is only provider to offer Ultra-high-field preclinical MRI systems of 11.7T and up to 18 Tesla pushing the envelope of what is currently possible in preclinical research.
Furthermore, Bruker is the only provider offering the MRI CryoProbe which provides a sensitivity boost compared to conventional room temperature coils making applications feasible which would not be possible without this solution.
With ParaVision 360,Bruker introduced a new software platform which seamlessly integrates multimodal workflows. ParaVision 360 offers convenient optimized procedures from animal positioning over scan programs, to data processing and management. This workflow concept is the cornerstone for data accuracy and reproducibility, leading to consistent quantification to facilitate a global discovery and validation process.

Q4 Reporter: In your opinion, what’s the trend in China PCI market in the near future?
Dr Jung: For MRI we see strong interest in field strength at high fields of 7 and 9.4 Tesla. There is also demand for even higher field strengths to complete for research areas where ultra-high fields add benefits (CEST, fMRI at high resolution, SWI, X-nuclei imaging, MR spectroscopy, etc.). The Chinese scientific imaging community shows a strong dynamic towards Ultra-high-field preclinical MRI which will open up unique possibilities for new, ground breaking discoveries in biomedical research. Brain research in non-human primates has also greatly pushed the development of large-bore animal MRI.
Furthermore, the growing demand for NMI and uCT products in China confirms the added value of multimodal imaging.
Q5 Reporter: To the Chinese specific market, what kind of strategic development plan will Bruker PCI make?
Dr Jung: The Chinese market is a key market for Bruker`s strategic development. We want to be the number one partner for the Chinese preclinical imaging community. We do not only offer the systems but also the support network from applications to maintenance and services.
We believe that China will become a most important place in clinical research and drug development if NHP therapeutic strategies will be validated in the future. Therefore, we are currently developing a new MRI instrument dedicated to non-human primate research to address this need.
We will further invest into the Chinese market. We already started this by hiring and training new personnel to specifically support the growing needs in China on all sides from sales and marketing up to after sales support.
