该研究证明精神压力等应激反应能够诱导糖皮质激素的迅速上升和Tsc22d3上调,抗体芯片检测结果显示所有重要免疫指标均被抑制,这会削弱抗癌疗法诱导的免疫监视作用,诱导免疫抑制降低化疗,免疫治疗等疗法的效率。此外在多种癌症包括消化道癌症、肺癌及黑色素瘤患者中也发现血浆皮质醇水平及TSC22D3在循环白细胞中的高表达,这与患者的消极情绪密切相关,并且预示较差的预后及生存率。因此糖皮质激素及其下游效应分子Tsc22d3可能作为“免疫检查点”,中和或阻断其作用可能有效地提高抗癌治疗的疗效。另一方面也提示人们需要多关注患者的精神障碍及调节、谨慎使用糖皮质激素类药物,为提高疗效提供了新的战略思路。

该研究首次揭示,组织特异性细胞的代谢重编程所引起的代谢物异常累积是银屑病等慢性炎症性疾病发生发展的重要“帮凶”,GSM-CAA-4000芯片结果表明表皮细胞的“多胺+自身核酸+短肽”复合物是激活皮肤免疫反应的“自身抗原”,并影响IL17相关细胞因子表达变化,促进病情发展。而靶向调节表皮细胞精氨酸代谢通路的异常有望成为治疗银屑病的全新策略。

研究团队发现血小板特异性Pten缺陷小鼠能够发生与年龄相关的淋巴细胞增生和自身免疫性疾病,通过使用小鼠细胞因子抗体芯片(AAM-CUST-8)比较了野生型及Pten缺陷型血小板上清中细胞因子的表达水平,发现PTEN缺陷能够过量产生Tfh相关的细胞因子。随后通过一系列实验证明Pten缺陷型血小板能够增强其与CD4+T细胞的相互作用,促进Tfh细胞分化,并能够浸润到淋巴组织并形成聚集体。
