研究背景:
高脂肪和高果糖(HFHF)饮食可驱动的脂肪毒性和糖毒性诱导肝损伤,从而促进树突状细胞活化,并产生主要的组织相容性复合体II类(MHC-II)免疫肽丘,富含来自细胞代谢、氧化磷酸化和应激反应的蛋白质的肽。
研究结果:
研究表明,证明了HFHF饮食驱动的脂肪毒性和糖毒性可引起代谢损伤可引起肝损伤,并促进由T和B细胞PDIA3表位驱动的致病性免疫自身反应性,参与免疫原性细胞死亡。
研究思路:
使用QAM-TH17-1分析对照和HFHF饮食小鼠的总肝细胞裂解液中是否含有炎症细胞因子。与对照组相比,HFHF喂养小鼠的pdia3反应细胞分泌的GZMB和多种炎症细胞因子(包括IL-17、IL-23、TNF和IFN-y)的量增加,表明HFHF喂养小鼠的肝脏中存在pdia3特异性T细胞,即使在没有免疫的情况下,对其同源PDIA3肽的增殖也会向功能性TH1和TH17型分化,并释放促炎细胞因子和细胞毒性分子。

本项研究向我们展示了新型巨噬细胞TAMEMs用于糖尿病伤口愈合的制备,通过细胞及体内实验验证了TAMEMs在小鼠糖尿病伤口中发挥的关键修复活性,包括调节炎症,刺激血管生成和促进增殖。其中利用RayBio 能同时半定量检测308种细胞因子的膜芯片(AAM-BLM-1)一次性地对TAMs – CM中的因子含量进行了检测,快速地锁定了重组蛋白培养基中关键蛋白的含量。

研究结果:
研究利用RayBio抗体芯片(GSH-ANG-2&GSH-ANG-3)检测出NPDR患者中血浆中ANG-1,PDGF-BB,TIMP-2和VEGFR2的血浆细胞因子增加,并且从血浆细胞因子中基于机器学习算法找到NPDR分类器,即从DM患者中进行对NPDR的预测。血浆细胞因子可能是NPDR的强风险标志物。
研究思路
①选取14例DM病人和14例NPDR病人,通过他们的血液样本进行蛋白芯片(GSH-ANG-2&GSH-ANG-3,共60个蛋白)实验,找到两组间表达的差异蛋白。
通过芯片实验结果每种蛋白的表达量绘制热图、火山图和PCA图等,加以限制条件(FC>4或者FC<0.25)进行筛选差异显著且差异倍数大的蛋白。
②选出的六个差异蛋白PDGF-BB, TIMP-1, TIMP-2, ANG-1, CXCL16和VEGFR2用验证集的大量样本(DM:115;NPDR:115)通过ELISA进行验证。
③用两种算法随机森林和lasso回归找到更为关键的三种蛋白(PDGF-BB、VEGFR2、ANG-1)进行后续的建模。
④通过多种模型(神经网络,逻辑回归,支持向量机,XGBOOST和随机森林)构建然后评估每个模型的准确率。最终找到表现最好的随机森林模型,预测的准确率达到85%。
