沙棘多糖抗运动性疲劳及抗氧化作用的研究

沙棘(Hippophae rhamnoides L.)作为一种常见的药食同源植物,具有较高营养价值和生物活性,常分布于我国西北、东北等地。沙棘多糖(polysaccharide from Hippophae rhamnoides,PBP)作为沙棘中最主要的活性成分具有抗氧化、抗疲劳、抑制肿瘤、降低血糖等药用价值。运动性疲劳会使机体产生大量的自由基和过氧化物,而抗氧化能力同样也会影响抗疲劳的效果。因此,本文主要研究沙棘多糖对长时间过量运动小鼠体力疲劳的缓解作用。小鼠分为空白组、PBP 低(100 mg/kg)、中(200 mg/kg)、高(400 mg/kg)剂量组,连续灌胃 15 d,记录力竭游泳时间,测定血乳酸(lactic acid,LA)、血清尿素氮(blood urea nitrogen,BUN)、肝脏中肝糖原(hepaticr glycogen,HG)含量,以及体外 DPPH 自由基、ABTS+自由基清除率和总还原能力。结果表明,与空白对照组相比,PBP 能够显著延长小鼠负重游泳时间(P<0.05),其中中剂量组游泳时间增加极显著(P<0.01)。PBP 中、高剂量组极显著(P<0.01)的降低了血清中 BUN、LA 含量,中剂量组极显著(P<0.01)的增加了肝脏中 HG 的储存量。体外抗氧化结果表明,沙棘多糖对 DPPH 自由基、ABTS+自由基都具有一定的清除能力,其IC50值分别为 0.81、2 mg/mL,当浓度为 1 mg/mL 时,总还原能力为 0.727。结果表明沙棘多糖具有一定的缓解体力疲劳、增强小鼠运动能力和抗氧化的作用。
Hippophae rhamnoides as a common medicine-food homologous plant, has high nutritional value and biological activity, and is often distributed in northwest and northeast China. Polysaccharide from Hippophae rhamnoides (PBP) , as the most important active ingredient in Hippophae rhamnoides, has the medicinal value of anti-oxidation, anti-fatigue, anti-tumor, reducing blood glucose and so on. Exercise fatigue would cause the body to produce a large number of free radicals and peroxide, meanwhile antioxidant capacity will also affect the effect of anti-fatigue. Therefore, this paper mainly studied the effect of polysaccharide from Hippophae rhamnoides on mitigating physical fatigue in mice with prolonged excessive exercise. The mice were divided into blank group, low (100 mg/kg), middle (200 mg/kg) and high (400 mg/kg) dose group of PBP, and fed continuous gavage for 15 d. Exhaustive swimming time was recorded. Then, the levels of blood lactic acid (LA), blood urea nitrogen (BUN) and the contents of glycogen. The results showed that, compared with the blank group, PBP significantly prolonged exhaustive swimming time (P<0.05). Exhaustive swimming time in middle PBP group increased significantly (P<0.01). And it significantly decreased the levels of BUN and LA in middle and high PBP groups. The level of HG in middle PBP group increased significantly (P<0.01). The antioxidant activity experiment in vitro showed that, the polysaccharide of Hippophae rhamnoides had certain scavenging ability to DPPH and ABTS+ free radical. And the level of IC50 was 0.81 and 2 mg/mL. When the concentration of polysaccharide of Hippophae rhamnoides was 1 mg/mL, the total reducing power was 0.727. Therefore, the polysaccharide of Hippophae rhamnoides can relieve physical fatigue, enhance the exercise ability of mice and have effect on antioxidant.
本实验结果表明 PBP 对自由基具有一定清除率,其中对 DPPH自由基清除能力较强,对 ABTS+自由基的清除能力较弱,其 IC50值分别为 0.812 mg/mL。PBP 的还原能力也随着浓度的增加逐渐增强。表明 PBP 具有良好的抗氧化活性。通过建立小鼠负重游泳模型,发现 PBP 能够有效延长小鼠负重游泳时间,增加肝脏中 HG 的储存量的同时降低血清中 BUN、LA 含量,实验结果证实 PBP 具有缓解体力疲劳、增强运动能力的作用。该实验结论与任薇、包晓伟及魏晨业等得出的结论一致虽然 PBP 浓度范围不同,但随着浓度的增加,都呈现上升的抗氧化趋势,尤其是对 DPPH自由基和ABTS+自由基清除效果较好。由于本实验抗氧化活性测定主要采用的是体外研究,具有操作简便、高效便于重复及实验周期短等特点,今后的研究可以采用细胞模型或动物实验,通过测定血浆、肝脏、心脏及脑组织超氧化物歧化酶、谷胱甘肽过氧化物酶和丙二醛等抗氧化指标的变化对 PBP 的抗氧化活性进行深入分析。为今后进一步发挥沙棘的资源优势,利用沙棘多糖开发保健食品和治疗药物等深加工产品提供理论基础。
